Born into a family of humble origin in Pozoblanco, Juan Gines de Sepúlveda went to school in Cordova, where he learned Latin and Greek, and at the age of 20 to Alcala de Henares University for three years (1510-13) before studying theology for two years at Colegio de Sigüenza. In May 1515, he left for the prestigious Colegio de San Clemente in Bologna with the support of the Chapter of the Cathedral of Toledo and Cardinal Cisneros.
